Nuclear import of HIV-1 -- optimizing siRNA techniques

In this project, the preliminary work for a study of nuclear import of HIV-1 in macrophages was conducted. RNA inhibition was used to prevent expression of karyopherins a1, a2, b2, and RCC1. siRNA techniques were tested and refined for each of the samples on HeLa cells by using real time RT-PCR to establish that mRNA inhibition occurred and performing Western blots to ascertain that specific cellular protein titers were lowered.
