Mutagenesis of the turnip crinkle virus

TCV-sv is a supervirulent form of turnip crinkle virus, capable of overcoming the hypersensitive response of the previously resistant Arabidopsis thaliana Di-17. Characterization of the virus has led to the identification of a 437 bp region which contains the mutations responsible for this supervirulence. Two non-silent mutations in this region are suspected of causing the supervirulence. The goal of this project is to create one of the mutations in wild-type TCV and to determine whether it alone is capable of causing systemic disease on Di-17.
