Mapping a mutation affecting the nervous system of C. elegans using PCR

The goal of this project was to determine the genetic location of nu385, a mutation in C. elegans thought to be involved in adhesion of neuronal processes during nervous system development. Genetic crosses were performed between strains carrying nu385 and strains carrying transposons or single nucleotide polymorphisms (SNPs) at known chromosomal locations. PCR and restriction digestion were used to observe the segregation patterns of these markers. nu385 was mapped to chromosome IV; linkage to the left arm of this chromosome is suspected. Additional testing is needed to prove this hypothesis.
