DOG BREED DIFFERENCES IN CYTOCHROME P-450 ISOFORMS IN LIVER MICROSOMES
PublicGreyhounds have been found to have decreased rates of clearance of the anesthetic propofol, in addition to a longer recovery period, as compared with other breeds of dog. Clearance of propofol involves hydroxylation, which has been shown to be mediated by cytochrome P-450 (CYP), a microsomal mixed-function oxidase system. Greyhound, beagle, and mixed-breed microsomal liver samples were quantitatively analyzed by the Western Blot technique for a variety of CYP isoforms. The amount of this drug-metabolizing enzyme present in greyhounds at least partially explains the pharmacokinetic differences.
