Conjugating a Neurotrophic Peptide to Blood-Brain-Barrier Carrier DHA Increases its Activity for Upregulating Therapeutic Enzymes SOD, Catalase and GPX

The purpose of this MQP was to extend analyses of the effect of various synthetic neurotrophic peptides on the upregulation of potentially therapeutic antioxidative enzymes in rat brains and hearts in vivo. Rats were treated with various doses of synthetic peptides representing different amino acid domains of neurotrophic factor Ependymin. Total cellular lysates were prepared from brain and heart, then the titer of specific antioxidative enzyme, SOD, catalase and glutathione peroxidase, was measured by immunoblot. The data indicate that attaching peptide 115-2 to DHA BBB-carrier increases its activity for upregulating SOD and GPX in the brain. Peptides 204-2, 204-16 and 115-40 were found to be inactive in the brain, but the latter two were active in the heart. Upregulation of GPX was optimal at 6-12 hours, and was transient. Drug 204-36 upregulated all 3 enzymes tested.
