Construction of a d-rR male genomic library

The Japanese medaka fish, Oryzias latipes, is fast becoming the most widely used fish model genetic system. The medaka fish is apt for genomic studies because of its small genome size (800 Mb), clearly defined sex chromosomes, resistance to disease, and their spontaneous and induced mutants provide a good model for sex determination and environmental carcinogenesis studies. The purpose of this project was to construct a medaka d-rR male genomic DNA library for performing future genre function experiments. The constructed library represents about 1,525,000 kb (an underestimation based on an assumed 10kb insert size) or about 1.9 medaka genomes. Therefore the library size coverage is sufficient to support further experimentation.
