Expression of CPx-ATPases in Arabidopsis thaliana

This project investigated the expression of CPx-type ATPases in Arabidopsis thaliana. RNA was isolated from Arabidopsis grown in the presence or absence of metals at different growth stages. Northern blotting was used to detect the presence of RAN1 and HMA4 mRNAs. The results confirmed transcriptional expression of both genes in Arabidopsis. His-tagged RAN1 was expressed in pBAD TOPO in Top10 E. coli cells. Expression was confirmed via western blotting. ATPase activity of the enzyme was measured in a membrane preparation.
