Evaluation of immunogold assays for amino acid neurotransmitters
PúblicoAgarose, gelatin, and PGA were assayed as potential matrices for adsorbing test solutions of amino acids, and gelatin was found to be the best candidate. Gelatin containing aspartate glutamate, or GABA was embedded in resin for ultrathin sectioning. Sections were treated with primary antibodies against amino acids and secondary gold-conjugated antibodies against the primaries, for examination by electron microscopy. The technique proved the reliability and specificity of antibodies against neurotransmitter candidates for applications in immunochemical studies using Transmission Electron Microscopy.
