Betafectin immunomodulator increases the activities of JNK and IKK kinases and increases the nuclear translocation of AP-1 in macrophage cells

The purpose of this MQP was to investigate intracellular changes that accompany the treatment of macrophage cells with a new immunomodulator drug. Substrate assays were performed to measure the activities of specific kinases representing a variety of signal transduction pathways. Immunoprecipitation Westerns were performed to monitor the movement of specific transcription factor antigens into the nucleus. EMSAs were performed to monitor increases in DNA binding affinity of transcription factor proteins. Our data indicate that the activities of JNK and IKK kinases increase, while p38 and Erk do not, that AP-1 increases in the nucleus, and AP-1 increases in DNA binding.
