Analysis of nutrient starvation of Pichia pastoris fermentation and the regulation of alcohol oxidase (AOX) as a simulation for expanded bed chromotograpy [sic]

As a continuation of research begun on the feasibility of coupling expanded bed chromatography (EBC) and yeast fermentation the issue of enzymatic regulation was investigated. To simulate the nutrient starvation conditions of the EBC process, fed batch fermentations of Pichia pastoris were deprived of nutrients for similar retention times to those of an EBC column. Analysis of pre and post starvation Alcohol Oxidase (AOX) regulation in the form of mRNA production was used as an indicator as to the effects of the starvation and the overall feasibility and validity of the coupled process.
