Detecting genetic structuring in Alosa pseudoharengus -- an evaluation of genetic markers

Alosa pseudoharengus a vulnerable species subject to numerous threats due to larval habitat loss. Under the Endangered Species Act, separate gene pools of a species are entitled to protection, provided their genetic distinctiveness is demonstrated. Several methods of measuring genetic structuring, namely microsatellite polymorphism, mtDNA markers, RFLP, RAPD, and AFLP are discussed and compared for efficiency and accuracy. In the end, the use of microsatellite markers is recommend, in conjunction with mtDNA tests to double check the results.
