Engineering a new turnip crinkle virus subgenomic RNA expressing p9

Turnip crinkle virus (TCV), an icosahedral positive-stranded RNA plant virus, expresses two movement proteins from same subgenomic RNA, (sgRNA) each with independent start codons. Owing to its internal initiation, the 3' product, p9, is expressed at undetectably low levels in infected cells. To induce higher levels of p9 expression and to allow for easier alteration of p9, a new TCV clone was created, where the 3' untranslated region of the TCV genome has been replaced with the p9 open reading frame driven by a TCV sgRNA promoter. Since there are indications that the 3' untranslated region has an effect on virulence, a set of control constructs has also been prepared to evaluate the effect of replacing the 3' untranslated region.
