Cellular Differentiation of the SH-SY5Y Neuroblastoma Cell Line into a Human Neuronal Cell Model

The neuroblastoma SH-SY5Y cell line can be differentiated into a mature, human neuronal phenotype through a variety of mechanisms. Despite there being many widely used protocols for differentiation, there are discrepancies between which methods select for specific neuronal subtypes. The purpose of this project was to create a differentiation model that can be used to investigate how expression levels of zinc transporters and chaperones change upon differentiation. To investigate cellular differentiation of the SH-SY5Y cell line, four different concentrations of retinoic acid were used to differentiate these cells. Morphological analysis confirmed cellular differentiation and total cell count revealed there was a correlation between the concentration of retinoic acid and the number of cells.
