First, to identify TGF- signaling pathways in PC3 cells and involvement of specific proteins such as PKC. Second, to discriminate between growth inhibitory and extracellular matrix inducing signaling pathways. This was accomplished using three experimental approaches: 1) Cell proliferation assays, 2) Signal transduction/gene expression assays, and 3) Western blot analysis identifying specific isoforms of down-regulated PKC. Although results of the first two approaches were inconclusive, cPKC was down regulated in response to PMA treatment.
