Insulin delivery by -PGA

-Polyglutamic Acid has the potential to be used as a slow release drug delivery system. PGA conjugated to insulin was administered to insulin starved Reuber H35 rat hepatoma cells, and by GSK-3 phosphorylation, was shown to mimic physiological insulin. Through time course and dose response experiments we determined that 10M PGA-insulin and 25 minutes showed the maximum GSK-3 phosphorylation. Receptor ligand crosslinking was performed to determine if insulin bound to the receptor retained glutamic acid residues from the polymer.
