Increasing the Efficiency of Monoclonal Antibody Production by Isolating Splenic B Cells in Advance of Hybridoma Fusion

open in viewerThe process of producing monoclonal antibodies using the hybridoma technology has been an arduous task for many years. In this study, we aim to improve the efficiency of monoclonal antibody production by isolating mature splenic B cells in advance of fusion using magnetic activated cell sorting (MACS) to select for cells expressing CD138, a cell marker present in mature B cells. Pre-isolating a population of cells expressing CD138+, but lacking CD19 and B220, resulted in a reduced number of cells required for fusion.
